HMB45
HMB45 (Human Melanoma Black 45) is a monoclonal antibody used in immunohistochemistry to detect melanocytic tumors, particularly melanoma. It targets a premelanosomal glycoprotein (gp100), which is expressed in melanocytic cells. HMB45 is most useful for identifying malignant melanoma, especially in distinguishing it from other tumors, and it can also highlight areas of melanocytic differentiation in ambiguous lesions. The staining pattern is typically cytoplasmic, with varying intensities depending on tumor activity. HMB45 is most reactive in immature, activated melanocytes. Limitations include potential false negatives in desmoplastic melanoma and non-specific staining in other neoplasms. For accurate diagnosis, HMB45 is often used alongside other markers like S-100 and Melan-A.4000 BDT
